Objective To investigate the effects of OGP10-14 and its derivatives G34C, G38I, G38K and G41F on bone of rats with ovariectomy-induced osteoporosis by bone histomorphometry and biomechanics. The active derivatives were screened out on the base of the experiments. Methods Sixty-four female Wistar rats, three month old, were randomly divided into 8 groups. The rats in group 1 to 7 were bilaterally ovariectomized (OVX) and only one group was Sham-operated. The groups 1 to 6 were treated by OGP10-14, G34C, G38I, G38K, G41F or estrogen(E) respectively after the ovariectomy while group 7 and 8 treated with vehicle along. The rats were sacrificed at 60 days after the ovariectomy. The tibia from each rat were reserved for bone histomorphometric study. The femora were separated apart and biomechanics were evaluated by three-points bending test. Results The Trabecular Bone Volume / Total Tissue Volume(TBV/TTV), Trabecular Bone Volume / Sponge Tissue Volume(TBV/SBV) and Mean Trabecular Plate Thickness(MTPT) were decreased in OVX group compared with Sham group while the Trabecular Osteoid Surface (TOS) and the...
