The Effects Of Osteogenic Growth Peptide C-terminal Pentapeptide Analogues OOJ, OOH And H88H On Bone Metabolism In OVX Rats

Objective To investigate the effects of osteogenic growth peptide C-terminal pentapeptide analogues(OOJ,OOH and H88H)on biochemical markers of bone tumover,bone histomorphometry,the ratio of ash weight to dry weight(AW/DW),bone mineral density(BMD)and biomechanical parameters in variectomized(OVX)rats.The relationships between these parameters were also studied.Methods sixty-nine female SD rats,three months old,were randomly divided into 8 groups.Group 1,low dose OOJ treated group(OOJL);group 2,high dose OOJ treated group(OOJ_H);group 3,low dose OOH treated group(OOHL);group 4,high dose OOH treated group(OOH_H);group5,low dose H88H treated group(H88HL);group 6,high dose H88H treated group(H88H_H);group 7,sham operated group(SHAM);group 8, ovariectomized group(OVX);Ovariectomy was carried out in group 1-7.After being injected subcutaneously with the above agents(Sham-operated and OVX rats being injected with phosphate buffered saline)once per day for 12 weeks,all rats were sacrificed. Uterus,kidney,liver,lumbar vertebrae(L1-5),bilateral tibiae and femurs were reserved from each rat.Serum alkaline phosphatase(ALP)and osteocalcin(BGP)was measured by biochemical analysis and radioimmunoassay respectively,The proximal tibial metaphysis was processed undecalcified for quantitive bone histomorphometry. The BMD was measured over the entire femur and lumbar vertebrae(L1-4)by dual energy X-ray absorptiometry(DEXA).Bone strength was measured by compression tests.The data were analyzed by SPSS 10.0.Results 1.Compared with group SHAM,the serum levels of ALP was significantly increased in OVX groups(P<0.01),there was an increased trend of the serum levels of BGP in OVX group,the serum levels of ALP and BGP were increased in H88H_L groups (P<0.05);2.the ratio of AW/DW was lower in OVX groups than which in group S-HAM, the difference was not statistical significant;Compared with group OVX,the ratioes of AW/DW in group OOJ_L,OOJ_H,OOH_L,H88H_L and H88H_H were significantly increased(P<0.01 or P<0.05),Compared with group SHAM,the ratio of AW/DW was increased in OOJ_H,OOHL and H88HL group(P<0.05);3,compared with group OVX, the BMD of femur and L1-4 were significantly increased in SHAM group(P<0.01), the BMD of L1-4 was significantly increased in each medication administration group (P<0.05);4.BV/TV,Tb.Th,Tb.N in group OVX were significantly lower than which in group SHAM(P<0.01),Tb.sp,OV/BV,OS/BS in group OVX were higher than that in group SHAM(P<0.01 or P<0.05),BV/TV and Tb.Th in group H88H_Land H88H_H were higher than in group OVX(P<0.01 or P<0.05),Tb.Th in group OOH_H was higher than in group OVX(P<0.05),there was an increased trend of OS/BS in group OOJ_L and H88H_L to be compared with group OVX,OS/BS was significant higher in group OOJ_L and H88H_L than in group SHAM(P<0.01).Histomorphometric datas showed that dL.s/BS in OVX group was higher compared with group SHAM(P<0.05),dL.s/BS was higher in group OOJ_L,OOH_L,OOH_H and H88H_L than in group SHAM(P<0.05),Compared with groups OVX and SHAM,MS/BS in group OOH_L,H88H_L and H88H_H were higher,BFRT in group OOJ_L,OOH_L,OOH_H,H88H_L and H88H_H were higher,the differences were not statistical significant;5.Compared with group OVX,there were an increased trend of maximal loading and elastic loading in group SHAM and each medication administration group except OOH_L group,there was an increased trend of energy absorption in each groups,maximal stress and elastic stress were higher in group SHAM than that in group OVX(P<0.05); 6.BV/TV and Tb.N showed a positive correlation with BMD.There wasn't significant relationship among bone ash content,BMD and biomechanical property.Convulsions The bone loss and bone quality decrease in OVX rats can be somewhat prevented by the analogues of osteogenic growth peptide C-terminal pentapeptide, It can promote BMD and improve the biomechanical characteristics.Beyond that,the result indicated that not only bone quantity,but also bone quality was important for the evaluation of the curative effects of any agents.
