Objective: To explore the changes of brain senescence-associated genes and the partial mechanisms of improving the learning and memory deficit in SAMP 10 by "YiQiTiaoXue, FuBenPeiYuan " acupuncture method. Methods: On the basis of prophase experiments which applying the cDNA Arraytechnique to sieve brain senescence-associated genes, Senescence Accelerated Mouse SAMP 10 and normal control SAMR1 were selected as model, the ability of spatial reference and working memory were determined by Morris Water Maze (MWM); applying the RT-PCR and DIG probed Northern blot techniques, the different expressions of Hsp84, Hsp86, NF-E2, Y box-binding protein 1 (YB-1), LRG47(Ifi1) genes were monitored in procerebrum, neocortex and hippocampus by 4 groups, viz.8-month SAMR1 control group, SAMP 10 control group, SAMP 10 "YiQffiaoXue, FuBenPeiYuan " acupuncture method group, SAMP 10 non-points group. Result: MWM indicated that:1 In spatial reference memory, comparing with SAMR1 control group, the abilities of learning acquisition, memory retaining and relearning of SAMP 10 control group decreased obviously. The courses of learning and memory were delayed . The abilities of managing strategies was absent and the edge and random pattern strategy was applied. While the spatial working memory of SAMP 10 decreased unremarkable.2 In spatial reference memory, comparing with SAMP 10 control group, the abilities of learning acquisition, memory retaining and relearning of SAMP10 "YiQiTiaoXue, FuBenPeiYuan" acupuncture method group increased markedly. The courses of learning and memory were shortened and the tendency pattern strategy was applied. Those of SAMP 10 non-points group increased unremarkable.Different expressions of 5 genes:1 In SAMP 10 control group, the expressions of Hsp84, Hsp86 and NF-E2, Y box-binding protein l(YB-l), LRG47(Ifil) were down regulated in procerebrum, neocortex and hippocampus.2 In SAMP 10 acupuncture method group, the expressions of Hsp84> Hsp86 and NF-E2, Y box-binding protein l(YB-l)^ LRG47(Ifil) were up regulated in procerebrum, neocortex and hippocampus.3 In SAMP 10 non-points group, the expression of LRG47(Ifil), NF-E2 in procerebrum and neocortex, YB-1 in procerebrum and hippocampus, Hsp86> Hsp84 in neocortex and hippocampus were up regulation unremarkable. The expressions of Hsp86, 84 in procerebrum and Ifil-. NF-E2 in hippocampus ,YB-1 in neocortex were up regulated..Conclusion:1 The ability of spatial reference memory in SAMP 10 was degraded. The acupuncture method of "YiQiTiaoXue, FuBenPeiYuari" increased markedly in the ability of learning acquisition, memory retaining and relearning, while the ability of spatial working memory in SAMP 10 was not obviously degraded.2 The abnormal expressions of 5 genes were concerned with learning and memory deficit in SAMP 10: Heat Shock Protein Hsp86 Hsp86 and Transcription Factors NF-E2^ YB-1 > LRG47(Ifil). Involved in the domain of cytoprotection, apoptosis, oxidative stress, erythrocyte series, cell proliferation and cell antibiosis immunity, the complexity of the mechanisms of senescence were well embodied.3 The possible mechanisms of improving the learning and memory abilities in SAMP 10 by "YiQiTiaoXue, FuBenPeiYuari' acupuncture method were those of increasing cytoprotection, suppressing apoptosis, resisting oxidative stress, improving the function of erythrocyte series, increasing cell proliferation and cell antibiosis immunity. These conclusions indicated that anti-aging of acupuncture be a sort of regulations of versatility and multistrata.4 The study indicated preliminarily that Transcription Factors NF-E2, YB-1 > LRG47(Ifil) were concerned with brain senescence and the deficit on learning and memory, while regulated optimumly by acupuncture.
