| Objective: To establish an optimized strategy of two-dimensional gel electrophoresis (2-DE) for proteome analysis of human pancreatic juice (PJ), screen tumor marker candidates and understand the pathogenesis of pancreatic cancer (PC) and chronic pancreatitis (CP). Methods: Effectiveness of different running conditions was compared according to their final results on 2-DE maps. Protein sports were identified by mass spectrometry (MS). RT-PCR were carried out for PC tissue specimen to validate proteins identified previously. Results: Seven differentially expressed proteins were identified. Two PC correlated proteins identified from previous PJ proteomic analysis were validated at gene expression level. Conclusions: 2-DE with MS is an ideal platform for differential proteomic analysis of human pancreatic juice. PSTI and TTR might be the potential tumor marker candidates. This proteomic study suggest some kind of correlation between inflammatory and tumor pathogenesis. |
