Objective: To explore the effect of tumor necrosis factor alpha on the kidney injury of rats with obstrutive jaundice and the underlying mechanism Methods: Sixty male Sprague-Dowley rats,weighting 200g to 250g were randomly assigned tofourgroups(n=15):control-group;sham-operation group;experiment one group and experiment two group.Except the control and the sham-operation group,the other two experiment group rendered jaundiced by doubly ligating the common bile duct.Glomerulus were isolated, the Glomerular mesangialCells (GMCs)werecultured.We measured cytosolic calcium concentration([Ca2+]i;the amount of nitric oxide(NO) and biological activity of iNOS .meanwhile,the changes of the biological activity of PC-PLD, apoptosis and mitochondrial membrane potential( △ψm) were observed.Results .Tumor necrosis factor alpha can induce GMCs apoptosis by way of ceramide.the mitochondrial membrane potential and the biological activity of PC-PLD were significantly decreased(p<0.05),the cytosolic calcium concentration and the amount of NO and iNOS were increased(P<0.05) Conclusion:Tumor necrosis factor alpha has significant effect on the kidneyinjury of the rats with obstructive jaundice ,ceramide mediate the influence oftumor necrosis factor alpha on the rats .
