PurposeObserving MFG-E8 plasma levels in benign and malignant obstructive jaundice patients; To study the expression of MFG-E8 in human cholangiocarcinoma and its correlation with clinicopathologic parameters.Methods1.Group: Randomly selecting 28 obstructive jaundice patients who were diagnosed with common bile duct stones as benign obstructive jaundice group(group B), the selected had no fever and cholangitis as well as other diseases; randomly selecting 21 obstructive jaundice patients who were diagnosed with cholangiocarcinoma as malignant obstructive jaundice group(group C); randomly selecting 25 healthy volunteers as the control group(group A).2.Collection : ?.Collecting healthy volunteers’ fasting blood in the morning as well as benign and malignant obstructive jaundice patients’ fasting blood in the first day morning after in hospital, keeping the supernatant after centrifugation,ELISA to detect MFG-E8 levels in plasma; ?.Collecting cholangiocarcinoma tissue after operation, Immunohistochemical study was performed to detect the expression of MFG-E8 in cholangiocarcinoma tissue; ?.Collecting cholangiocarcinoma patients’ clinicopathologic parameters.Results1.Compared with the control group(group A), MFG-E8 plasma levels decreased significantly in benign obstructive jaundice group(group B)(P<0.05);MFG-E8 plasma levels increased significantly in malignant obstructive jaundice group(group C)(P<0.05);MFG-E8 plasma levels in benign and malignant obstructive jaundice group is Significant differences, malignant obstructive jaundice group(group C) is higher than benign obstructive jaundice group(group B)(P<0.05).2.The expression of MFG-E8 in cholangiocarcinoma tissue is higher than Tissues adjacent to cancer(P<0.005);The intensive expression of MFG-E8 in cholangiocarcinoma tissue was positively correlated to histologicgrade(P<0.05),clinical stage(P<0.005) and metastasis(P<0.005).Conclusions:1.The MFG-E8 plasma level was significantly decreased in benign obstructive jaundice patients caused by common bile duct stones, which could have impact on removal of large apoptotic cells after obstructive jaundice, may be one of mechanism for multiple organ dysfunction.2.The intensive expression of MFG-E8 was found in cholangiocarcinoma tissues.and plasma, and was correlated to histologic grade,clinical stage and metastasis. which suggested that MFG-E8 is a promising biomarker for the diagnosis and prognosis target of cholangiocarcinoma.3.The MFG-E8 expression may be a useful biomarker to distinguish benign and malignant obstructive jaundice.
