Objective: To establish an animal model of perineural dissemination of adenoid cystic carcinoma (ACC), study the characteristics of perineural dissemination in ACC via this animal model and identify the role of dorsal root ganglion (DRG) cells and nerve growth factor (NGF) on the perineural dissemination of ACC in vitro. Methods: Tumor pieces of ACC were transplant into infraorbital area of nude mice to establish a perineural dissemination model of ACC in animal;The characteristics of perineural dissemination in ACC were studied via animal model;The role of dorsal root ganglion cells and different concentrations of hβNGF on proliferation, apoptosis, adhesion, motor ability and invasion of ACC-2 cells were studied through experiments in vitro. Tca-8113 cell were taken as control group. Results: The animal model of perineural dissemination of adenoid cystic carcinoma was established successfully. ACC were observed to grow consecutively at the raritas interspace of nerves and not to invade the nerve fibers;hβNGF at certain concentrations can promote the proliferation, adhesion and invasion ability of ACC-2 cells. DRG cell conditioned medium can also promoto the invasion ability of ACC-2 cells. Conclusion: The ACC cells closed to nerves may be regulated by DRG cells and hβNGF, and the proliferation, adhesion and invasion ability of ACC-2 cells were then promoted. It may result in the perineural dissemination of ACC.
