Objective: To study the expression of telomerase in keloids and its rule and significance in the formation and development of keloids. Methods :The immunohistochemistry method was used to detect the expression of hTERT, bcl-2 and p53. in keloid lesions. The PCR-ELISA technique was used to assay the telomerase activation of KFb. The expression of "hTERT and bcl-2 mRNA in the KFb were detected by using semiquantitative RT-PCR analysis. KFb in culture medium in vitro were given IFNa-2b .The proliferation of the fibroblasts were measured by cell count and MTT assay, the changes of telomerase activity of KFb were detected by PCR-ELISA method, the apoptosis was analysed by FCM , and the expression of hTERT and bcl-2 mRNA were obsevered with RT-PCR.Results: The expression of human telomerase were strong in keloid, The expression of hTERT in keloids had significant correlation with the expression of bcl-2 and p53. IFNa-2b could inhibit the growth of keloid fibroblasts and down-regulated its telomerase activity significantly. Conclusions: telomerease activation could be a key factor in the development of keloid and had significant correlation with the high expression of tumor gene(p53,bcl-2) in keloid. These findings suggest that telomerease may play a important role in the pathogenesis of keloid. That some drugs inhibited telomerase activity in keloid fibroblasts is an new pathway that may play a key role in the anti- keloid therapy.
