Objective:To investigate the expression of cyclinD1,P16 and cyclinA1 in thefibroblasts of keloids and hypertrophic scars, explore the relationbetween the genesis of two different kinds of scars。Methods:12 cases of keloid and 11 cases of hypertrophic scar were obtained astwo experimental groups. Normal skin tissues were used for thecontrol group and the expression of cyclinD1,p16 and cyclinA1 werescreened in hypertrophic scars and keloids by immunohistochemicalSP method.Positive expression rate were statistical analysed. Results: (1)The expression rates of the three proteins in normal skin fibroblastsare negative(positive rate < 10%). (2) In the group of keloid, positive expression rates of cyclinD1,P16 and cyclinA1 are 91.7%,33.3% and 75.0%; positive expression rates of cyclinD1,P16 andcyclinA1 in hypertrophic scar are 45.5%, 81.8% and 27.3%. (3)Compared with control group, the expression rates of cyclinD1 andcyclinA1 in keloid group and P16's expression rates in hypertrophicscar are higher, and there are significant difference(P<0.01);Theexpression rates of P16 in keloid and cyclinD1, cyclinA1 inhypertrophic scar are also higher than in normal skin, but there arenot remarkable difference(P>0.05). (4)Compared with hypertrophicscar group, the expression rates of cyclinD1 and cyclinA1 in keloidgroup are higher, and there are significant difference(P<0.05); however, the expression of P16 in keloid is lower than that inhypertrophic scar, and there is significant difference(P<0.05).Conclusion : The expressions of cyclinD1,p16 and cyclinA1 in the fibroblasts ofkeloids and hypertrophic scars are all higher than that in normalskin.High expressions of cyclinD1 and cyclinA1 as regulatoryfactors in G1 and S phases and the relatively low levelexpression of P16 as negative regulatory factor in G1 phase leadto the continued growth of keloid and the tendency of selfrestrictionin hypertrophic scars. CyclinD1,p16 and cyclinA1 arein disequilibrium which may result in the different developmentof different kinds of pathological scars.
