The Detection And Clinical Significance Of Active Telomerase In Pathological Scar | | Posted on:2005-11-01 | Degree:Master | Type:Thesis | | Country:China | Candidate:W H Xie | Full Text:PDF | | GTID:2144360125965295 | Subject:Surgery | | Abstract/Summary: | PDF Full Text Request | | Objective: pathological scar after skin wound is the one of the most intractable issue for the aesthetic plastic surgery,but the mechanism of the pathological scar formation is still unclear.Telomerase activity is the base of cellular immortality.This study determined telomerase activity in pathological scar, in order to evaluate the role of the telomerase activity in the formation of pathological scar; To evaluate the difference between hypertrophic scar and keloid; To provide the experiment evidence for the mechanism and prevention of the pathological scar formation.Methods: The study material is 46 sample of pathological scar and nomal skin by clinic acquired,including 13 keloid,10 hypertrophic scar,23 nomal skin. Specimen ground in liquid nitrogen in after cut breake fully in earthen bowl.Telomerase activity was determined by telomeric repeat amplification protocol(TRAP)in pathological scar and nomal skin.The products were analyzed inpolyacrylamide gelelectrophoresis with silver staining and the presence of a ladder with 6 base pairs increments was considered as positive result.SMMC-7721 cells were used as positive control,and the extracts from the positive control cells were heated in 650C for min as negative control. the tissue section of pathology scar was observed in light microscope after HE dyeing.The TLMA were determined respectively,the statistic method is exact probability in fourfold data.Result: TLMA activity was positive in 7 of 13 keloids(53.85%); 1 of 10 hypertrophic scar(10%); none of the whole normal skin group(0%). The positive ratio in the keloid group is higher than others (p<0.05). But there were no pathological difference between the keloid group and hypertrophic scar group in microscopy,their elementary structure unit is collagen nod, as well as in electron microscopy. Conclusion: 1.Telomerase activity was detected in keloids(7/13),which indicates high level of telomerase exprssion may be related to the pathgensis of the keloid.2.Telomerase activity was detected in hypertrophic scar (1/10),which indicates high level of telomerase exprssion never related to the pathgensis of the hyperplasia scar.3.Telomerase activity was detected,which was in taken of a laboratory of difference scar and hypertrophic scar index. 4.Tissue structural of keloid and hypertrophic scar was similars.
