Objective: To observe the effect of Guyanding to the proliferation of cultured human chondrocytes in vitro, and the level of expression of type II collagen and iNOS under influence of IL-1.And discuss the mechanism of cartilage protection with Guyanding recipe.Method: Human chondrocytes were obtained from femoral head cartilage came from femoral neck fracture THA surgery and cultured in vitro. The Guyanding was filtrated by grug sera and used in the culture solution. All chondrocytes were divided into four groups: control group, IL-1 group, IL~1 and L-NAME group, IL-1 and Guyanding group. The proliferation of chondrocytes was comprared between the control group and Guyanding group by MTT method. Moreover, the level of of expression of iNOS and type II collagen was compared between the four group mentioned above, by ELISA and ROPCR method. Result: Chondrocytes were high proliferation in Guyanding group than in control group. In IL-1 group, the expression of iNOS was significantly higher than in control group, but the level of Type II collagen expression was significantly lower than in control group. After the iNOS inhibitator L-NAME was added, the level of expression of iNOS was almost as same as in the control group, at the mean time the expression of Type II collagen was significantly higher than in the IL-1 group. The same phenomenon was observed in the Guyanding group.Conclusion: Guyanding can stimulate the proliferation of human chondrocytes cultured in vitro, and inhibit the expression of iNOS, increasethe expression of Type II collagen under the influence of IL-1. As a conclusion based on the phenomenon mentioned above , Guyanding has protection effect to chondrocytes and the cartilage matrix.
