The p53 gene is a kind of tumor suppressor gene. Its main function is inhibiting cell proliferation, inducing apoptosis and leading to the cell differentiation. When the DNA is damaged, the wild type p53 gene arrests the cell in G1 phase of the cell cycle. If the damagement cannot be repaired, the p53 will induce the cell to enter the apoptosis process in order to maintain the balance between the cell proliferation and the cell differentiation. If the mutation or loss of p53 occurs, the induction of the apoptosis cannot be initiated and the genome instability and integrity can be destroyed, resulting in the cell proliferation in a unrestricted way. The inactivation of the p53 gene by missense mutation or deletion is the most common genetic alteration in human cancers.
