Endothelium-derived contracting substance (EDCF and A â…¡ etc.) and relaxing substance (PGI2 and EDRF etc. ) remain interactive and keep dynamic equilibrium in normal. Vasomotion of visceral vessel may directly influence the blood pressure variation, but reports concerning the endothelium-derived factors changed in mesenteric artery during hypertension were lacking. These changes may play an important role in hypertension. Until now there is no any systematically evidence indicated how changes and what mechanisms of endothelium-derived factors, cGMP, NOS and [Ca2+] i in the endothelium-dependent relaxation (EDR) of mesenteric artery in Stroke - prone spontaneous hypertensive rats (SHRSP) and spontaneous hypertensive rats (SHR) are. So the purpose of the present study was by using microbioassay technique,RIA, the method of NADPH -Diaphorase (Nd) histochemistry and measurement of intracellular calcium by the Fura- 2/AM microflurosecent technique in single cultured vascular smooth muscle cells to systematically investigate the changes of endothelium...
