Objective: To investigate the relationship between telomeraseexpression, MN expression and the carcinogenesis of cervix , and the possibility of use them as markers to monitoring and to evaluate the natural history of CIN.Methods: Use the telomere repeat amplification protocol(TRAP)assay to detect the telomerase activity of cervical lesion in cervical scraping samples and cervical biopsy tissue.The fragment of MN cDNA was obtained from RNA extraction of HeLa cell by RT-PCR. Then through the technology of gene recombinant, we obtained the MN-Pï¿¡T22B(+) plasmid. Inducting MN expression , and then purificating fusion protein, the protein was injected into BalB/C mouse to prepare anti-MN mouse polyclonal antibody. Immunohistochemical staining was conducted for ervical tissues and the cercvical exfoliative cells of cervical lesons by utilization of the anti-MN polyclonal antibody. Result: 62.5%,60%,82.4% and 61.9% cervical scraping sampleexhibited telomerase activity in CIN- I ,II, III and cervical cancer, respectively, compared 50%,45%,96.43%and 95.23% in cervical biopsy tissue .In contrast,20% and 26.67% in cervical scraping samples and...
