Esophageal cancer (EC) is one of the most frequent killers among malignant diseases, ranking forth in China and seventh in the world, with five-year survival rate less than 10%. Geographic variation in incidence of esophageal cancer is very striking. There are several districts with high incidence of this disease in China. Screening of precancer and early stage cancer of esophagus is an important approach for reducing the incidence rate and death rate. To explore esophageal cancer-associated autoantibodies as biomarkers for the screen of esophageal precancer, a modified SEREX method was applied for screening, isolation and identification of esophageal precancerous antigens. A protein microarray technique with these antigens was established to serological screening of the esophageal precancerous patients from high-risk population.Part One. Serological identification and characterization of esophageal precancer associated antigensA cDNA expression library of 1×10~6 primary clones was prepared from 14 mixed tissues of esophagus, including one moderate differentiation squamous cell carcinoma (ESCC) and 13 dysphasia(DY). The library was immunoscreened with IgM antibodies from three sera pools, including a pool of 13 autologous DY, a pool of 42 allogenic ESCC and a pool of 32 allogenic DY. Seventy-five positive clones were...
