Objective: 1. To study and compare anticoagulation effects of heparin, low molecular weight heparin, antithrombin, recombinant hirudin on disseminated intravascular coagulation. 2. To compare fibrinolytic activity, uPA receptor and annexin II between various leukemia cell lines and to investigate the alterations of these indices in APL cell line in the presence of all trans retinoic acid.Methods: 1. Therapeutical effects of different decoagulants on experimental animals'DIC. (Dconstructing animal model. A model of disseminated intravascular coagulation was established by injecting lipopolysaccharide(2.5mg/kg) in rabbits' peripheral vein within 10 minutes in two doses. The interval time of administration was 1h. (2)Thirty rabbits were randomly divided into five groups(control group, heparin group, low molecular weight heparin group, antithrombin group and recombinant hirudin group). Each group contained six rabbits. They were injected with normal saline, heparin(100IU/kg), low molecular weight heparin(100U/kg), antithrombin(100U/kg) and recombinant hirudin(100IU/kg) at the same time as lipopolysaccharide injected. Activated partial thromboplastin time(APTT), thrombin time(TT), fibrinogen, fibrin degradation products, antithrombin , platelet counts, white cell counts, hemoglobin were determined before and 2,4,6hs after iv lipopolysaccharide. At the end of experiment, the rabbits were killed and the heart, liver, kidney, spleen and lung tissues were separated and fixed with 4% paraformaldehyde. After HE staining, the histological changes were observed under light microscope. 2. Studying fibrinolytic activity of various leukemia cell lines in vitro. (1)Cell culture. NB4, K562, Jurkat, Raji cells were propagated in RPMI 1640 medium supplemented with 15% heat-inactivated bovine serum, while SHI-1 cells were propagated in IMDM medium...
