Objective:To investigate the effect of hyperbaric oxygen(HBO) on the symptoms, the changes of vital signs,peripheral blood cells,clotting factor indicators,pathology of the lung and kidney in rabbits with disseminated intravascular coagulation(DIC).Methods:1.Animal model constructing.A model of DIC was established by injecting a single bolus of 100μg/kg lipopolysaccharide(LPS) in rabbits via ear vein, time of injection was more than 10 minutes.2.Twenty two rabbits were randomly assigned into three groups,group A(n=6) received normal saline(30ml,bid),group B (n=8) and group C(n=8) received additional oxygen therapy at atmospheric pressure and 2.5 absolute atmospheric pressure respectively.The general condition of rabbits, vital signs,white blood counts(WBC),platelet counts,activated partial thromboplastin time(APTT),prothrombin time(PT) and the level of plasma fibrinogen(FIB) were determinated before and 6,24,72h after injection of LPS.Then,the rabbits were sacrificed 72h later to observe pathological changes in the lung and kidney under microscope.Results:After injection of LPS,rabbits in group A revealed apathy,dyspnea, cyanosis,and oliguria.Rates of respiratory and heart were accelerated.Rectal temperature was decreased.Counts of WBC and platelet were decreased significantly. The level of FIB was declined.APTT and PT were prolonged.Micro thrombus, hemorrhagic foci and infiltration of phiogocyte could be observed in the lung and kidney.Compared with group A,the general condition and vital signs of rabbits in group B did not improve obviously,counts of WBC and platelet,the level of FIB,PT were not change(P>0.05),only APTT was shortened(P<0.05).Hemorrhagic foci and infiltration of phlogocyte also could be observed in group B.The general condition and vital signs of rabbits in group C was improved noticeably.Counts of WBC and platelet, and the level of FIB were significantly higher than that in A group and B group (P<0.01).APTT and PT were shorter than that in A group and B group(P<0.01, P<0.05).Moreover,no hemorrhagic foci and inflammation were observed in the lung and kidney of rabbits in group C.Conclusions:We concluded that 1) HBO treatment could block coagulation cascade formation and platelet activation,then reduce the consumption of clotting factor and platelet,alleviate micro thrombus and bleeding in essential organs.2) HBO therapy could reduce the destruction of white blood cells,lessen inflammation,increase the supply of oxygen and blood in important organs,and improved the symptoms of DIC markedly.
