Section I :EFFECTS OF REACTIVE OXYGEN SPECIES ON SPERM MOTILITY, SPERM BEMBRANE FUNCTION AND SPERM MITOCHONDRIAL MEMBRANE POTENTIALObjective: To study the machanism of normal sperm damage indcuced by reactive oxygen species(ROS):evaluate the effect of ROS on sperm motility,viability,sperm membrane integrity, lipid peroxidation in sperm membrane and acrosome integrity. At the same time, explore the changes of mitochondrial membrane potential (MMP) in sperms of oxidative stess model with fluerescent JC-1.Methods: Spermatozoas with normal physiological functions selected from semen samples by swimming-up method were regarded as normal sperm models. ROS were generatede by ferrous sulfate (0.02mmol/L) and hydrogen peroxide system at dosage of 0.05,0.25,0.5,1,5mmol/ml. Sperm VCL, VSL, VAP, BCF, ALH, LIN, STR, MAD were assessed by computer assisted sperm analysis (CASA).
