Infection of Human cytomegalovirus (HCMV) is universal and almost no symptom appears in immunocompetitive persons, but it may result in severe diseases or even death in immunocompromised individuals. With the advent of specific immunotherapy and preemptive antiviral therapy, the incidence of HCMV associated diseases and death in the receptors of transplantation decreased dramatically. Accordingly, it is extremely important to explore the measures used in the laboratory diagnosis and adoptive immunotherapy for those who are susceptible to HCMV associated diseases for the previous interference. This will be helpful to decrease the abnormality and severe neural damage from HCMV intrauterine infection as well as increase the achievement of transplantation. On account of this purpose, three parts are involved in this investigation. 1. Prokaryotic expression of pp150 of HCMV and its antigenic analysisBackground Up to now, the detection of specific IgM antibody is the major measure for the laboratory diagnosis of HCMV active infection. The HCMV antigens used in the ELISA kit usually come from virion of cytomegalovirus prepared in fibroblasts. Because of the cross reaction between the antigens of HCMV and HSV (human simplex herpes virus) as well as between the HCMV and membrane of human cells, it is difficult to standardize the antigens and as a result, default reports are often presented in serological diagnosis.
