PART 1STUDY ON THE PREPARATION OF POLYBUTYLCYANOACRYLATE NANOPARTICLES LOADED HYPOCRELLIN B AND ITS PHARMACOKINETIC/PHARMACODYNAMIC IN TUMOR BEARING FISCHER344 RATSObjective:To develop a nanoparticle drug delivery system carrying the second generation photosensitizer Hypocrellin B for the application of photodynamic therapy on ovarian cancer. Methods:The emulsification evaporation method was used to prepare HB-PBCA-NP. According to the results of the effects of different variables, the uniform experimental design was applied to optimize the technology of preparation. The properties of nanoparticle as diameter and size distribution were observed by transmission electric microscope (TEM) and laser dispersive analyzing apparatus for granularity.The encapsulation efficiency and drug loading capacity were assayed by ultra centrifugation at low temperature and HPLC .NuTu-19, a cell line derived from a poorly differentiated adenocarcinoma of Fischer 344 rat , was cultured in vitro. 1 × 10~6 cells were injected intraperitoneally to female Fischer 344 rats to to develop a spontaneously arising, nonimmunogenic experimental animal model of epithelial ovarian cancer.
