For the moment , cancer is threatening peoples' health more seriously, searching for the safe and effective anti-tumor components from natural medical materials has become an important part of studying anti-tumor drugs in the world.Qingtiankui comes from the rhizome and whole grass of Nerviliae Fordii (Hance) Schltr in orchidaceae plants. Its actions include moistening the lung and relieving cough, cleaning heat and detoxifying fire poison, breaking up congealed blood and stoping pain. It is primarily used for hemotysis induced by pulmonary tuberculosis, lung heat cough, stomatitis, sore throat goiter, scrofula , traumatic injuries, et al. In mordern research, Qingtiankui can relieve Nasopharyngeal Carcinoma(NPC) clinical symptoms. But no chemical constituents or pharmacological studies of qingtiankui have ever been reported. Its anti-tumor activity also need to be further proved. The fore-experiments of its chemical components showed that it contained triterpenoids, steroids, organic acid, amino acid and flavonoids, et al.The alcoholic extract obtained from the dried grass was divided into four portions including petroleum ether extract, ethyl acetate extract, n-Butyl alcohel extract and methanol extract for the first time. Anti-tumor experiment in vitro was done by MTT assay to different extracts on human leukemia L1210 and P388D1, uterine carcinoma Hela, human gastric adenocarcinoma SGC7901 , B16 Melanoma cell, neurornas NG108-15, Human hepatoma Hele7404. The results showed that petroleum ether extract and ethyl acetate extract both had some anti-tumor effects in vitro. To the effective parts preliminary selected in vitro, the MTD experiments of mice were done. The results showed the mice were in good condition during drugs, the MTD dose of mice was 18.0g/kg to petroleum ether extract of Qingtiankui, and the MTD dose of mice was 14.4g/kg to ethyl acetate extract of Qingtiankui. To the effective parts preliminary selected in vitro, anti-tumor experiments in vivo were also done by S180 -mice and H22-mice models. Petroleum ether extract and ethyl acetate extract of Qingtiankui also both had obvious anti-tumor effects to S180-mice and H22-mice, and could prolong H22-mice life. Both could improve the immunoloregulation of mice, too.Effective parts were seperatcd and purified by chromatographic methods. Pury...
