Proteomics Research On Synovitis Of Collagen-induced Arthritis (CIA) Rat Treated With Bizhongxiao Decoction (BZXD)

Objective:(1) To provide useful and new clues for elucidating the pathogenesis of rheumatoid arthritis by exploring the proteomic research on synovitis of rat with collagen-induced arthritis ;(2) To elucidate the mechanism of bizhongxiao decoction (BZXD) in treating rheumatoid arthritis by analyzing the differential expression proteins of synovitis of collagen induced arthritis (CIA) rat treated with BZXD.Methods:(1) The experiment arthritis rat model was established by injected subcutaneous of bovine â…¡ collagen(B â…¡C) and complete Freunds adjuv-ant(CFA).The CIA rats were divided into 2 groups randomly,the Bizhongxiao decoction treatment group(BZXE) group, n=30) drenched with BZXD and the CIA model group(model group, n=30), drenched with the same dose of double distilled water(DDW). Normal control group (Normal group, n=10) drank water freely.(2)General condition, weight, arthritis index score, degree of paw swelling and pathology was observed in 3 groups. The arthritis index(AI) of the rats was examined in CIA group and BZXD group on 10th day, 15th day, 20th day, 25th day, 30th day, 35th day, 40th day and 45th day respectively. The weight and ankle thickness of rats was measured in three groups every week. The pathomorphology of ankle was observed on 25th day , 35th day ,45th day.(3) The total proteins of synovial tissue of rat joint at different stages in normal,model and BZXD group were seperated by two-dimensional gel electrophoresis(2-DE)respectively.
