Background and Objective:Graft-versus-host disease (GVHD) and graft rejection , the significantcomplications of bone marrow transplantation (BMT),limit the using of BMT to spread. Natural killer (NK) cells exert a variety of biologic characteristics. We will study the function and mechanism of NK cells in prevention of GVHD and graft rejection. First, we must establish a method of purifying and identifying NK cells and a mice GVHD BMT model. And then, using the purifying NK cells and the model, we will examine whether allogeneic NK cells would reduce GVHD and enhance engraftment after allogeneic BMT in mice and further analyze the relation of these function with the contents of host total antigen-presenting-cells (APC) or dentritic cells(DCs) , host mature APC or DCs ,and transforming growth factor-β1(TGF-β1) .
