Objective:To explore the possible protective mechanism with controlled release bFGF—PLGA Microspheres from two aspects of the cell apoptosis and nerve regeneration of injuried spinal cord in rats. Methods: 1. To elaborate bFGF—PLGA Microspheres(PLGA microspheres) by multiple emulsion encapsulative method and to observe its morphology with electron scan microscope. To measure the drug loadingrate and drug embedding rate of the microspheres. To study the release property of the microspheres in vitro and to prepare the freeze-drying powder for using. 2. 24 female SD rats were randomly divided into 2 groups.False traum group: laminectomy of T8 only were operated in 12 rats, nerval traum group : T8 segment spinal cord were exposed and were injured with modified Allen' WD method(20g × 2.5cm) in 12 rats. After SCI(Spinal cord injury), morphological changes were observed, SEP and BBB locomotor rating scale were examed. Then,a small hole on the dura matter shortly near the posterior middle vessels was acquired by microtechnique, spinal fluid flowed...
