Objective 1. To establish and study the effect of pancreaticoduodenal transplantation model in allogenic rats.2. To study immune tolerance induced by human AdCTLA-4IgG after pancreaticoduodenal transplantation in rats. Methods 1. The diabetes Wistar rats were induced by intravenous administration of streptozotocin ( STZ) in a single dose of 60mg/ kg via penis dorsal vein. 2. Pancreaticoduodenal transplantations were performed with the SD rats as donors and the diabete Wistar rats as recipients. During the operations, vein anastomoses were performed with cuff technique, artery anastomoses were performed with consecutive suture without overturn of the grafts, the grafts were hung in the air to avoiding warm ischemia,and was protected by plastic patches to gain heat preservation and to avoid water losing, tempreture recovery was gained with the help of warm water as soon as the blood supply of it was regained. 3. The SD pancreaticoduodenal grafts transfected with human AdCTLA-4IgG via intraarterial ex vivo perfusion were transplanted into diabetes Wistar rats of transfer group .
