| Background:Rejection after kidney transplantation is one of the vital influencing factors which affected the survival rate of transplant recipient,long-term application of immunosuppressant has its inevitable disadvantages.Therefore,inducing immune suspression of transplanted kidney successfully is an important topic of the transplantation class.Human leucocyte antigen G(HLA-G)is a factors of immune tolerance that play an extremely important role in building Materno-fetal immune tolerance and normal pregnancy,as well in organ transplantation,expression of HLA-G of postoperative patients has been proved to reduce the incidence of rejection.high expression of HLA-G molecules,is expected to become a new approach to induce immune tolerance with assistant of genetic engineering some day.Objective:In this study,we transplanted the HLA-G gene into the rat transpla nted kidney and transplanted the transplanted kidney into the recipient rat.By detecting the pathological changes of the transplanted kidney and the expressio n of the HLA-G gene,The effect of transplanted kidney transfected HLA-G ge ne on renal allograft rejection after transplantation was analyzed,and a powerf ul research tool was provided for the basic experimental study of HLA-G gene-induced immune tolerance of transplanted kidney.Methods:The experimental group was transfected with lentivirus containing HLA-G gene.The negative control group was transfected with lentivirus without HLA-G gene,and the blank control group was perfusion The transplanted kidneys were transplanted into the recipient renal arteriovenous vein by the establishment of the arteriovenous cuffs,and the transplanted kidney bladder valve was sutured to the recipient bladder wall to complete the rat kidney transplantation The establishment of the model.After the success of the model,the rats were randomly selected three weeks after the model,and the rats were randomly divided into three groups.The rats were sacrificed and the transplanted kidneys were removed.The rats were sacrificed and removed.The The expression of HLA-G gene was analyzed by HE staining and immunohistochemistry and Western blot analysis.The immunological rejection of transfected HLA-G gene was analyzed by HE staining.influences.Results:A total of 30 rat kidney transplant models were established,3 died,90%success rate,2 of them died of bleeding,one died of urinary fistula.The rejection of the control group was significantly lower than that of the control group,and the expression of HLA-G gene was detected by immunohistochemistry and Western blot analysis.The expression of HLA-G gene was not expressed in the control group HLA-G gene,while the experimental group expressed HLA-G gene,and with the practice of prolonged,increased expression.Conclusion:In this study,the rat model of renal transplantation was established by establishing renal artery and vein cuff method,which effectively reduced the operation time and simplified the complexity of operation,which provided new ideas for the generalization of kidney transplantation model.At the same time,the experiment also found that transfection of HLA-G gene can effectively reduce the occurrence of acute immune rejection after transplantation,delay the immune rejection,for transfection of HLA-G gene induced renal transplantation immune tolerance experimental study laid the foundation. |
