Purposes Collagen is a major extracellular matrix component in prolif-erative vitreoretinopathy (PVR), but the contribution of collagen to the pathophysiology of PVR is not known, we measured the procollagen III (PC III ) levels in the vitreous of patients with PVR, to determine the potential role of collagen III in the pathogenesis of PVR. We also investigated the effects of the cytokines on the collagen synthesis by human retinal pigment epithelial (HRPE) cells that are thought to play a important role in PVR, to study the modulation mechanisms of the collagen synthesis involved in PVR, and to evaluate a possible adjunctive therapy for PVR by IFN - γ. Methods PC III levels were measured by radioimmunoassay in vitreous of patients with PVR, and the relationship between PC III levels and some factors closely related to PVR were analyzed with Logistic regression analysis. The effects of TGF - β , IL - 1β and IFN - γ on the collagen synthesis in cultured HRPE were examined by ~3H — proline incorporation assay and by indirect im-munofluorescence reaction with anti — human type I collagen and type III and type IV collagen antibodies. The steady — state amounts of mRNAs for type I and III and IV collagen were analysed by dot - blot hybridization, by using specific cDNA probes for these molecules.
