To Explore The Mechanism Of Acupuncture And Moxibustion In Treating Ischemic Stroke From Inherent Neural Stem Cells

Ischemic stroke is a common disease in clinic It has been proved in clinic practice that acupuncture is effective on ischemic stroke. However, researchers paid most attention to neural protection mechanism in the past when they studied the influence of acupuncture on ischemic stroke in lab, little about neurogenesis after ischemia. our experiment is initially compared moxibustion and acupuncture therapy , which do affect neurogenesis after ischemic stroke.Objective: From inherent neural stem cells ,to explain the possible mechanism of acupuncture and moxibustion therapy on ischemic stroke . we studied the effects of acupuncture and moxibustion on neural stem cell proliferation and differentiation, also compared the differences between acupuncture and moxibustion.Methods : A local brain ischemia model was made by occluding middle cerebral artery with an intraluminal suture. Neurological deficit score and pathological transformation in all groups were compared to estimate ischemic injury after treaded 24 day. To evaluate the roles of acupuncture, moxibustion in neurogenesis after ischemia, expression of nestin and BrdU was calculated as markers of neural stem cell by immunohistochemistry. Content of NGF is measured by enzyme linked immunosorbent assay. In polymerase chain reaction,. expression of nestin was calculated as markers of neural stem cell, also NSE incorporation and GFAP expression as markers of proliferation anddifferentiation. Results:1.HE dye indicate: Compared with model group, the injury ofacupuncture and moxibustion group is slighter in tissue and cell.2.Neurological deficit score indicate: Compared with modelgroup, the recovery of acupuncture and moxibustion group is rapider.3.Enzy linked immunosorbent assay indicate: Compared acupuncture and moxibustion group with model group, the content of NGF is plentiful.4.1mmunohistochemistry indicate: The expression of Nestin and BrdU ependyma and dentate gyrus were elevated after ischemia. Compared acupuncture and moxibustion group with model group, the expression of BrdU and Nestin is higher.5.Polymerase chain reaction indicate: Compared acupuncture and moxibustion group with model group, the expression of Nestin> NSE is rising in mRNA, the expression of GFAP is decline in mRNAConclusion:l.The acupuncture and moxibustion could increase the creation of NGF in MCAO rat's brain.2.The acupuncture (Electro-acupuncture) could increase proliferation and differentiation of neural stem cell in ependyma and dentate gyrus.3.The moxibustion could increase proliferation and differentiation of neural stem cell in ependyma and dentate gyrus.4.Our experiment indicate effect of moxibustion is stronger than acupuncture after ischemia stroke.5.It is one of the possible mechanism of acupuncture and moxibustion therapy on ischemic stroke that acupuncture and moxibustion could increase proliferation and differentiation of neural stem cell.
