Objective: From inherent neural stem cells, we explained the possible mechanism of acupuncture and moxibustion therapy on ischemic stroke. We studied the effects of acupuncture and electroacupuncture on neural stem cell proliferation and differentiation also compared the differences between acupuncture and electroacupuncture.Methods: A local brain ischemia model was made by occluding middle cerebral artery with an intraluminal suture. Neurological deficit scores and pathological transformation in all groups were compared to estimate ischemic injury after treated 28 day. To evaluate the roles of acupuncture and moxibustion in neurogenesis after ischemia, expression of BrdU and Nestin were calculated as markers of neural stem cell by immunohistochemistry. Content of NGF was measured by enzyme linked immunosorbent assay. In polymerase chain reaction, expression of GFAP was calculated as markers of neural stem cell, also Nestin incorporation and NSE expression as markers of proliferation and differentiation.Results: 1. Neurological deficit score indicate: Compared with model group, the recovery of acupuncture group and electroacupuncture group were rapider. 2. HE dye indicates: Compared with model group, the injury of acupuncture group and electroacupuncture group were slighter in tissue and cell. 3. Immunohistochemistry indicates: The expression of BrdU and Nestin in dentate gyrus and subventricular zone was elevated after ischemia. Compared acupuncture group and electroacupuncture group with model group, the expression of BrdU and Nestin were higher. 4. Enzyme linked immunosorbent assay indicate: Compared acupuncture group and electroacupuncture group with model group, the content of NGF was plentiful. 5. Reverse transcription polymerase chain reaction indicates: Compared acupuncture group and electroacupuncture group with model group, the expression of GFAP was decline in mRNA; the expression of Nestin and NSE were rising in mRNA.Conclusion: 1. The acupuncture and electroacupuncture could promote the recovery of MCAO rat function. 2. The acupuncture and electroacupuncture could increase the creation of NGF in MCAO rat s brain. 3. The acupuncture and electroacupuncture could increase proliferation and differentiation of neural stem cell in dentate gyrus and subventricular zone. 4. Our experiment indicated the effect of electroacupuncture was stronger than acupuncture after ischemia stroke. 5. It was one of the possible mechanisms of acupuncture and moxibustion therapy on ischemic stroke that acupuncture and moxibustion could increase proliferation and differentiation of neural stem cell.
