Up to now , it is commonly accepted that psoriasis is a kind of diseases with the abnormal cellular immunity under the background of polygenic inheritance. The disease is characterized by epidermal hyperproliferation, abnormal differentiation of keratinocytes and skin inflammation. Retinoids have been shown to be highly effective in the treatment of psoriasis and mediate their biologic effects through binding to nuclear receptors known as retinoic acid receptors (RARs) or retinoid X receptors (RXRs). In the normal epidermis, the expression of RARγ/RXR α is the most abundant. Although the ligands of the retinoid receptor superfamily and its derivatives are the first-line reagent to treat psoriasis, the specific function of retinoids on psoriasis is still obscure, which hold back the development and utilization of the new drugs relating to retinoids.The objectives of this study are: (1) to study whether the level of expression of the retinoid receptor is related to the pathogenesis of the psoriasis or not. (2 ) to make clear the therapeutic mechanism of retinoids to treat psoriasis. (3) to find out at which target cell is aimed directly by retinoids as these drugs are used for psoriasis.In this study, the methods to be established were as follows: RT-PCR, immunohistochemistry of tissue and cell, the technique of culturing or subculturing of colo-16 cell in vitro; moreover, in the study, we optimized the methods of extracting RNA from the epidermis, conserving RNA and RNA electrophoresis.
