In this study, two new human pancreatic adenocarcinoma cell lines of PC-7 and PC-48, were established by adding soybean trypsin inhibitor to the culture medium.Up to now, the cell lines have been cultured continuously more than 150 (PC-7) and 50 (PC-48) passages in vitro. They were proved to be human pancreatic adenocarcinoma cell lines by morphological and immunocytochemical studies with anti-keratin and anti-CEA antisera. They shared common characteristics of short population doubling-time,high mitotic indices, and low diploid, abnormal chromosomal karyotypes.By immunocytochemical staining and Northern blot hybridization, expression of EGF and EGFR at proteins and mRNAs levels was detected in PC-7 and PC-48 cell lines, and compete-inhibit combination teBt of 125I-EGF with EGF receptor demonstrated the presence of EGFR on PC-7 cell membrane. Growing slowly in low serum or serum-free medium,was accorded with the characteristic of reduced serum-dependence of PC-7.The effects of exogenous EGF, anti-EGF and anti-EGF receptor antisera treatment on the growth of PC-7 cell line in low-serum medium were: (1) EGF stimulated significantly the cell growth and hyperplaBia of PC-7, with high cell-count and high 3H-TdR incorporation rate as compared with the control; (2) Anti-EGF antiserum inhibited significantly the cell growth of PC-7, with low cell-count and low 3H-TdR incorporation rate.
