Purpose: to evaluate the effects of human interleukin 2 gene transfer on the biological behaviors of ovarian cancer cell line SKOV3 and the possibility of IL-2 modified tumor cells as a newer tumor vaccine. Method: hIL-2 cDNA was amplified with PCR and inserted into the retrovirus vector pLXSN. After packing, hIL-2 cDNA was transduced into ovarian cancer cell line SKOV3. IL-2 activity was detected. The morphologic changes, proliferation, tumorigenesis, adhesive ability, immunity, chemotherapeutic sensitivity and oncogene expression was assayed. Results: The recombined vector pIL-2SN was constructed with the insertion of IL-2 cDNA PCR product. 8 G418 resistant clone with high IL-2 activity (16~318U/10~6cells/24hours) in the culture supernatants was isolated. IL-2 activity was detected by Immunohistochemistry and RT-PCR also. Microscope and morphologic studies showed that SKOV3/IL-2 cell was more homogeneous than SKOV3 and SKOV3/Neo cells, with increased cell acreage but decreased nucleus/plasma ratio. The cytoplasm vacuolization and the karyotin condensing of SKOV3/IL-2 cell was showed by electronic microscope. The frequency of karyokinesis in SKOV3/IL-2 cell was less than SKOV3 and SKOV3/Neo cells. The growth curve of SKOV3/IL-2 cell was more flat than the control's. The doubling time was prolonged. H~3-TdR uptaking, and PCNA expression detection indicated that the hIL-2 transfer has decreased the proliferation of SKOV3 cells. The cell cycle analysis revealed that the G0/G1 phase was markedly increased in the hIL-2 gene modified cells. The clone forming and soft-agar clone forming efficiency decreased from 52% and 32% of parental cells to 0.7% and 2.4% of hIL-2 modified cells respectively. The cell adhesion test showed that hIL-2 modified cells adhense less effectively to plastic substance, collagen IV and matrigel.
