The growth factor-like effects of gastrointestinal (GI) peptide hormones on cell proliferation, differention and tumor genesis have become a hot point of research recently. Bombesin is a 14 amino acid GI peptide with growth factor-like effects. Studies in recent years showed that bombesin has trophic effect on many kinds of cell lines and tissues, especially on tumor cell lines. The mechanism regulating cell growth and proliferation by bombesin is not very clear. Moreover, the relationship between bombesin or other GI hormones and cell cycle regulating factors have not been reported. The aim of this study was to investigate the growth-stimulating effect of bombesin on immortalized human gastric epithelial cell line GES-1, the regulatory effect of bombesin on cell cycle regulating factor cyclin D1 and CDK4, so as to explain the mechanism of growth regulatory effect of bombesin.Methods: Using thiazol Blue( MTT ) as yhe parameter of cell growth, the growth- stimulating effects of bombesin, bombesin antagonist ([D-phe~6] BN (6-13) methyl ester )on the GES-1 cells were studied. The GES-1 cells were treated with bombesin at different concentrations, alone or together with antagonist, and cyclin D1 positive cell were checked by flow cytometry. Western blotting and enhanced chemo- luminescence (ECL) were performed to identify CDK4 protein expression in GES-1 cells treated by bombesin.
