Objective To investigate the probable related genes and ERK signal transduction pathway in the pathogenesis of malignant melanoma (MM).Method In the first part of our study, we extracted total RNA from four MM samples and normal controls, and isolated their mRNA. Then the mRNA was reversely transcripted into cDNA, which at the same time mixed into 33P. After that, the production was hybridized with membranes of cDNA microarry, which contained 2000 human genes each, to study samples gene expression profiling. Finally, we used quantitative real-time RT-PCR to confirm down-regulation of two co-differentiated expressed genes we interested in. In the second part of our study, we used immunohistochemical methods on slides from paraffin-embedded tissues of MMs, normal skins, ordinary nevi and Spitz nevi, to assessed expression of some ERK pathway related proteins, which were ERK1, ERK2 and Cyclin D1.Results There were totally 151 genes showed more than 2 times different expression by cDNA microarray hybridization. 4.7-6.15%, 0.75-1.4% and 0.45-0.5% genes on the membranes showed 2, 5 and 10 times different expression respectively.
