Liver fibrosis is the essential process to liver cirrhosis. Liver fibrosis will be reversed and cured if it can be diagnosed and treated at the early stage. Our group focuses on the study of induced expression of metallothionein by selenium and its anti-fibrosis effect. The experimental results showed that metallothionein can be induced significantly by Org-Selenium and has considerable effect on the protection of liver fibrosis.AIM Evaluation of inducing effect of Se on the expression of MT; extraction, purification and identification of MT-1/2 induced by Org-Se; establishment of liver fibrosis model in animal and establishment of protein expression profile in serum and liver tissue; application of liver damage related bio-markers to evaluate the protection effect of MT induced by Org-Selenium; clinical analysis of therapeutic anti-fibrosis effect of Org-Se ; establishment of protein finger print of liver fibrosis patient; at the same time, search of bio-marker related with liver fibrosis.METHODS (1) Comparison of inducing effect of Org-se and Zinc on the expression of MT at transcription and translation level by various methods, such as SELDI-TOF-MS and RT-PCR; Using classic method to abstract, purify and identify MT-1/2 induced by Org-Se and zinc; purity of MT was verified by SDS-PAGE; molecular weight of MT-1/2 was identified by MALDI-TOF-MS.
