Objective TO investigate whether 17β-estradia (E2) can prevent retinopathy of prematurity(ROP) and its possible mechanism by observing the influence of physiological level' estradial on bovine retinal vascular endothelial cells (BRECs) under different oxygen conditions and the effect on retinal neovascularization in ROP of SD rat.Methods MRNA expression of vascular endothelial growth factor (VEGF), hypoxia-inducible factor-1α (HIF-1α) and endothelial nitric oxide synthase (eNOS) were studied by reverse transcription polymerase chain reaction (RT-PCR) in BRECs, with E2( 10-12,10-10,108mol/L)or phosphate-buffered saline (PBS),under normoxiaand hypoxia ,after 8hours and 24hours.In a rat ROP model, effects of E2 were evaluated by daily subcutaneous injecting either 17β-estradiol (E2, 1 μg/rat) in vehicle (dissolved in ethanol and diluted in 0.1 mL/rat of PBS) or vehicle alone (PBS) during hyperoxia [ (75±2) % O2]from postnatal day 1 to 7 (P1-7) and/or after exposure to room air(P8-14,or P1-14). The developmental condition of rat was observed every day. At P7,P14,three rats were chose to be anesthetized randomly in every group to obtain retinal flatmounts by using a perfusion of ink,in order to to study the retinal vascular pattern; The eyes were enucleated after the rats were executed ,mRNA expression of VEGF, HIF-1α, eNOS in the retina was studied by RT-PCR; At P14, the eye-tissue -sections were obtained and stained with hematoxylin-eosin to analyze the extraretinal neovascularization, or...
