Chapter I Expression and clinical significance of apoptosis associated proteins Bcl-2 in CholangiocarcinomaOBJECTIVE: To study the expression of apoptosis-associated protein Bcl-2 in normal bile duct and cholangiocarcinoma. To analyze the relationship between the positive rate of Bcl-2 expression and the clinical pathological parameters in cholangiocacinoma, and between the positive rate of Bcl-2 expression and the post-operative survival rate in cholangiocacinoma. To evaluate the significance of Bcl-2 protein expression in predicting prognosis and in the genesis and development of cholangiocarcinoma. METHODS: The expression of Bcl-2 protein was detected by SP immunohistochemical staining in paraffin sections from 36 cases of human cholangiocarcinoma and 10 normal bile duct specimen. Log-rank test and Kaplan-Meirer method were used in comparing survival curve and the survival rate.The relationship between the positive rate of Bcl-2 expression and the clinical pathological parameters in cholangiocacinoma, and between the positive rate of Bcl-2 expression and the post-operative survival rate in cholangiocacinoma were analyzed by statistical software package SSPS13.0. RESULTS: The positive rate of Bcl-2 protein expression in cholangiocarcinoma tissues was 47.2%( 17/36), while that in normal bile duct tissues was 10.0% (1/10) with significant difference between them (p<0.05).The positive rate of Bcl-2 protein expression in poorly-differentiated cholangiocarcinoma tissues was 75.0%(6/8), while that in mediate-differentiated cholangiocarcinoma tissues was 46.2 %(6/13) and in well-differentiated tissues was 33.3%(5/15) with significant difference between them (P<0.05). No significant difference between the positive rate of Bcl-2 protein expressions and other clinical and pathological parameters such as the tumor size and lymphnode metastasis was found (P>0.05).
