Enamel is the hardest, most highly mineralized tissue in the vertebrate body.The exact role of extracellular matrix protein in controlling the synthesis of tooth enamel remains unclear.The amelogenin protein are the principal secreted products of cells known as ameloblasts, which are found only in the enamel organs of developing teeth , and it has been estimated that amelogenins constitute 90% of the secreted enamel organic matrix.Many roles have been proposed for amelogenins, including crystal nuclestion, regulation of growth of individual mineral crystals, action as an ion reservoir or calcium buffer during hydroxyapatite formation, and provision of support and protection for the growing crystals.The amelogenin gene cloning is a popular problem in the world. But previous attempts to use mature rat and European embryo cDNA library have proven difficult and expend money and time. We used the molecular biological techniques to clone and sequence the rat and Chinese embryo, and express in E.coli and purify.Then an anti-amelogenin antibody is prepared to proceed the Western blot and immunohistochemical study.This paper includes following 4 parts:一, Cloning and sequencing of Rat and Chinese embryo...
