The early diagnosis is still the major means to elongate the survival rate of the patients with gastric and colorectal carcinomas. Tumor markers provide a new approach to the early diagnosis of cancers. Mucins' family which has been discovered recently plays an important role in the progress of the tumor. So far, there is no report about the correlation between the serum levels of mucins and gastric and colorectal carcinomas.Objective: Establish mucins' protein chip and test the serum levels of mucinl, mucin2 and micin5ac in gastric and colorectal carcinomas patients and health control group. Investigate the expression pattern of mucins' protein in cancer and control groups and evaluate the preliminary application of the mucins' protein chip in the diagnosis of gastric and colorectal carcinomas.Methods: Mucins' protein chip was established with monoclone and multiclone antibodies and applied to measure the serum levels of MUC1, MUC2 and MUC5AC in 30 gastric cancer patients and 30 colorectal cancer patients and 30 healthy adultsas control, using the method of immuno-fluorescence.Results: The serum mucins' and CEA levels of gastric cancer patients were significantly higher than that of the healthy ones( MUC1 5.85 ± 2.57vs 3.65 ± 1.82 p<0.01, MUC2 4.46 ± 2.47vs2.73 ± 1.63p<0.01, M UC5AC 5.07 ± 3.24vs 3.16 ± 1.57 p<0.05,CEA 8.52 ± 3.36vs 5.36 ± 2.58, p<0.05) .The positive expression of the MUC1was related with the gastric cancer's TNM stage(p<0.01). The positive expression of the MUC2 and MUC5AC had not relationship with the gastric cancer's stage(p > 0.05). The positive expression of the mucins was related with the gastric cancer's differentiation.The positive rate of CA19-9, CA15-3, CA-125, CA72-4 in gastric cancer patients was 16.7%, 6.6%, 20.0%, 23.3% respectively, while the positive rate of MUC1,MUC2and MUC5ACwas 73.3%, 73.3%, 66.6% respectively. The sensibility and specificity were both higher in mucins when used to diagnose. The sensibility and specificity of MUC1 was 81.5%, 75.8% respectively. The sensibility and specificity of combined mucins (at least 2 in 3 mucins were positive) was 96.0%, 82.9%. The serum mucins' and CEA levels of colorectal cancer patients were significantly higher than that of the healthy ones(MUC1 5.25 ±2.37vs3.65 ± 1.82 p<0.01, MUC2 4.28 ± 2.14vs2.73 ± 1.63p<0.01, MUC5AC 5.04 ± 3.20vs3.16 ± 1.57 p<0.05, CEA 8.58 ± 3.48vs5.36 ± 2.58p<0.05).The positive expression of the mucins was not related with the cancer's stage(p > 0.05). The positive expression of the mucins was related with the cancer's differentiation.The positive rate of CA19-9, CA15-3, CA-125, CA72-4 in colorectal cancer patients was 20.0%, 6.6%, 6.6%,16.7% respectively, while the positive rate of MUCl,MUC2and MUC5ACwas 76.6%, 63.3%, 60.0% respectively. The sensibility and specificity were both higher in mucins when used to diagnose. The sensibility and specificity of MUC1 was 82.1%, 78.1% respectively. The sensibility and specificity of combined mucins (at least 2 in 3 mucins were positive) was. 96.2%, 85.3%Conclusion: Serum levels of MUC1, MUC2, MUC5AC and CEA in patients with gastric and colorectal cancer were higher than in healthy adults. The positive expression of the MUC1was related with the gastric cancer's TNM stage, the positive rate was higher in advanced gastric cancer than in early stage gastric cancer. The positive rate of MUC1, MUC2, MUC5AC was up when tumor's differentiation was low. But in colorectal cancer,the positive rate of MUC2 was up when tumor's differentiation was high. Comparing with CA19-9, CA15-3, CA-125, CA72-4, MUC1, MUC2, MUC5AC had higher sensibility and specificity in the diagnosis of tumor. When combined mucins were used in diagnosis,the sensibility and specificity was much higher. The establishment of protein chip has provided a new approach to early diagnose gastric and colorectal cancer.
