Actinidia kolomikta as a plant of Actinidiaceae is mainly distributed in Northeast region and Changbai Mountain of China, North Korea, Japan and Russia. The fresh fruit of the plant has activity in cure Vitamin C lack and in physical strong,the fresh or dry leaves of the plant also used as drugs.According to the literature,Flavonoid, leucocyanidin, Kaempferol, lactone, saponin were found in the leaves of Actinidia kolomikta,High contents of Vitamin C was determined in its mature fruits, 0.03% alkaloid was determined in its roots.Flavonoid Constituents as Kaempferide -7-O-rhamnoside and Kaempferide -7-O-(rhamnosyl) -3–O - rutinoside were seprateded and identified from the leaves of Actinidia kolomikta By Chang Xiaoli .For the purpose of the further study on the chemical constituents and bioactivities of Actinidia kolomikta, the extraction, separation and determination of the leaves of Actinidia kolomikta from Chanbai Mountain were preformed. Furthermore, the bioactivities of total flavonoids on cardio-cerebro vascular diseasis were studied also.7 Flavonoid compounds were separated and identified by 1HNMR, 13CNMR, 1H-1H COSY, HMQC, HMBC, NOESY, MS and IR as Kaempferide -3 -O-rutinoside(compoundI),Kaempferide-7-O-(4″-O-acetylrhamnosyl)-3-O-glucoside(compoundII),Kaempferide-7-O-(4″-O-acetylrhamnosyl)-3-O-rutinoside (com -poundIII),Kaempferol-3-O-β-D-glucoside(compoundIV),Kaempferol-3-O-rutinoside(compoundV),Kaempferide-3-O-β-D-glucoside(compoundVI) and Kaem -pferide -7 -O-rhamnosyl–3 -O-rutinoside (compound VII).Except compound VII, All the compounds are obtained from Actinidia kolomikta for the first time. Compound I~III are new compounds .To determine Kaempferide- 7-O- (4″- O- acetylrhamnosyl) -3–O - rutinoside (compound III) in the leaves of Actinidia kolomikta and the contents with different growth periods ,the RP-HPLC was used. The separation was performed on ZORBAX Extend C18 column (250 mm×4.6 mm,5μm), using 51% methanol and 49% water mixture as mobile phase with the rate of 1.2 mL?min-1 at 25℃, the wavelength for measurement was 267 nm. The results showed that the content of Kaempferide-7-O-(4″-O-acetylrhamnosyl)-3-O-rutinoside in the leaves of Actinidia kolomikta were variety in different growth periods. It could reach their highest content in the middle ten days of June,then, the content would decrease in the middle ten days of July slightly, it could reach their lowest content in the middle ten days of August. Thus, The optimal collective date for the leavesof Actinidia kolomikta are the middle ten days of June.The bioactivities of the total flavonoids extracted from the leaves of Actinidia kolomikta has studied in this thesis, the results showed that it could reduce the infarck bulk after lacking blood, the provent rose on cerebra damage submitted from blood lacking in a dose-dependent manner. Moreover, The the total flavonoids can reduce MIS amount of acute myocardial infarct rats evidently, decrease the activities of CK,LDH and AST in serum of myocardial infarct rats with the FFA amount increasing;The extracts decrease LPO content, increase SOD and GSH-Px levels in plasma; Also, the extracts can decrease ET and Ang II levels,increase PGI2 level and PGI2/TGX2 ,decease the visicosity of plasma and blood.To sum up, the total flavonoids extracted from the leaves of Actinidia kolomikta has bioactivities in the cardiac muscle and cerabal cell peotecting in blood lacking , the thrombus formation preventing. So, It's hopeful that the leaves of Actinidia kolomikta will be developed a new drug in curing cardio-cerebro vascular diseasis vessel .
