This study use chemical, biochemical, Confocal laser technology and quantitative PCR and other advanced means and methods of detection to explore viability, the cell structure changes and the regulation of apoptosis of PC12 cells injured by Amyloidβ-protein and affected by the cerebrospinal fluid-amyloid with Decoction of Rehamanniae. Aβ25-35 PC12 cell injury model, the current international standards of AD model, was established and the neurotoxicity effect of apoptosis induced by Aβ25-35 was confirmed.The results showed:1, Decoction of Rehamanniae can effectively improve survival and viability of PC12 cells, and the pathological changes of the structure and function induced by Aβ25-352, Decoction of Rehamanniae can significantly reduce the excessive reaction with the stimulation by the Aβon PC12 cells and the Protein expression of APP, thereby inhibit the PC12 cells injury.3, Decoction of Rehamanniae can raise the Aβexpression level of NOS and NMDAR, and inhibit gene expression of ICE mRNA to inhibit apoptosis production.4, Decoction of Rehamanniae can improve the culture medium free calcium, inhibit calcium influx, enhance calcium-binding protein expression, inhibit calcium overload and Aβthe memb- rane structure.
