Alzheimer's disease (AD) is a kind of neurodegenerative disease that causes progressive cognitive and behavior deterioration in the elderly over 65. As the life span of human beings increasing, the incidence of AD is increasingly high,which is a heavy burden to AD patients' families and the society. It is important for us to find ways to protect and treat AD.We adopt the methods of invigorating the kidney and replenishing essence, dispelling phlegm and quickening blood to cure AD. Decoction of Rehmanniae (DR) is in keeping with the pathogenesis characteristic of root vacuity and tip repletion.DR follows the principle of treating the asthenia-syndrome by therapy of invigoration and treating the sthenia-syndrome by purgation. So it is a effective prescription of protecting and treating AD.In our experiments, the model ras were administered D-gal intraperitioneal injection and β-Amyloid protein fragment1-40 into hippocampus. Mean well we used the technique of optical lens, electron microscope, immunohistochemical method, biochemistry and molecular biology.We studied the mechanisms that DR pretect and treat AD from the aspects of apoptosis, inflammation injury, oxidative injury , cholinergic neuron injury and the behavior experiments at cellar and molecular level. The results can be expressed:1.DR improves the ability of active and passive avoidance response and spatial probe in the behavior experiment to improve the rat's ability of learning and memory.2.DR pretects hippocampal neurons from the damage of free radicals by improving the activity of antioxidative enzyme CAT and decreasing MAO in rats brain.3. DR increases the expression of NF-kβ and hsp70 gene and decrease the expression of caspase3 gene to inhibiting the apoptosis of hippocampal neurons injured by AP.4.DR increases the expression of SYN of cortical and hippocampal neurons by improving the injury of cholinergic neuron to recovery the function of synaptic transmission.5.DR decreases the expression of IL-1α and TNF-α to inhibit the activity of glia cell and inflammatory injury of hippocamal neurons.
