Objective: (1) The aim of this study is to investigate the effects ofinhibitor HNP-1 transfection on proliferation and tumor growth of humannasopharyngeal carcinoma cell line HNE-1. (2) The expression levels ofa-defensin-1 in malignant epithelial cells were examined for investigatingthe role ofα-defensin-1 in nasopharyngeal carcinoma patients.Methods: (1) Expression of HNP-1 wasn't demonstrated by reversetranscriptase polymerase chain reaction in FINE-1 cell line. HNE-1 cells weretransfected with HNP-1 by liposome, and the cytotoxic effect was detectedby MTT, PI. In vivo tumor growth test was performed in female BALB/cnude mice inoculated with HNE-1 cells of transfected pcDNA3.1(+)-HNP-1plasmid and only pcDNA3.1(+) plasmid. The therapeutic effect of HNP-1was evaluated in the inoculated tumors. Expression ofα-defensin-1 wasdetected in implanted tumor tissues by immunohistochemical stain. (2)Nasopharyngeal carcinoma tissue was taken from 68 outpatients and theexpression levels ofα-defensin-1 in malignant epithelial cells were examined.64 of 68 cases patients were nasopharyngeal poorly differentiated carcinoma,4 cases were well-differentiated carcinoma. According to the standard of clinic stage of UICC(2002):â… stage 10cases,â…¡stage 20cases,â…¢stage30cases,â…£stage 8cases. To investigate the correlation of expression humanα-defensin-1 with age, gender, clinical stage, immunohistochemical stain wasadopted.Results: (1) HNP-1 transfection significantly inhibited the proliferationof HNE-1 cells. In vivo study showed tumor volume was significantlysmaller in HNP-1 transfection group than that in control group (P<0.05).Immunohistochemical stain showed the expression of a-defensin-1 wasincreased in HNP-1 transfection group.(2) Nasopharyngeal carcinoma cell of 26 of 68 cases patients werepatchy stained by immunohistochemical stain. Among the positive group:male 20, female 6, average age 49.95,â… stage 4cases,â…¡stage 12cases,â…¢stage 14cases,â…£stage 6cases. Carcinoma cell of 4 caseswell-differentiated carcinoma were patchy stained; negative group: male30, female 12, average age 50.03,â… stage 6cases,â…¡stage8cases,â…¢stage16cases,â…£stage 2cases.Conclusion: (1) HNP-1 transfection can inhibit the proliferation ofHNE-1 cells as well as tumor growth in nude mice. (2) No obviouscorrelation of expression of humanα-defensin-1 with age, gender, clinicalstage, but it seems correlateion with grade of tumor. This study can'tconfirmed the correlation between the exepression of humanα-defensin-1with prognosis of patients.
