[Objective]To Provide the theoretical evidence for photodynamic diagnosis of nasopharyngeal carcinoma by applied photosensitizer(photocarcinorin PsD-007)on the experiment on human nasopharyngeal cancer xenograft in nude mice models .[Methods]The CNE-2 cells were transplanted into the subcutaneous tissues of 22 nude mice for building murine nasopharyngeal carcinoma models. According to the dose of PsD-007 which was injected into nude mice through tail veins, mice were divide into group A and B randomly. The cancer tissues and control tissues were exposed to Ar+ laser light with following parameters: wavelength 514.5nm, exposed time 60s, energy density 1.08 J/cm2. Meanwhile, the fluorescent image was obtained. Tumor tissues and mouse nasopharynx tissues were acquired and sectioned, stained with haematoxylin. The result were observed and analysed.[Results]1. Spss 13.0 software was used for statistical analysis. The result of analysis through Chi-square Test showed p value <0.05. It implied that there was statistical difference between group A and B.2. Pathological results of tumor samples proved poorly differentiated squamous cell carcinoma. of 20 cases showed positive results with laser-photodynamic diagnosis, the other 2 cases showed negative.3. Under high magnification, the nasopharyngeal mucosa which was irradiated by Ar+ laser light were observed. No acute inflammatory response or tissue injury was detected. [Conclusions]1. Laser-photodynamic diagnosis has high sensitivity in diagnosis of nasopharyngeal carcinoma. With proper pseudo-color threshold for the fluorescent images, the better performance in distinguishing the tumor tissues and normal tissues could be achieved.2. The corresponding laser parameters and photosensitizer in this study were nondestructive to tumor tissues and control tissues. That is, they are safe for living body.
