The present work dealt with the immobilization of papain on mesoporous molecular Sieves of the M41S family with the aid of glutaraldehyde and the main contents could be divided into three parts:Ⅰ. MCM-41 as well as MCM-48, two of members of M41S family,was prepared and characterized, whose peak pore diameters of these two obtained materials were all between 6 and 7 nm.Ⅱ. Papain immobilization on MCM-41 with the aid of glutaraldehyde was investigated, and the characteristics of this immobilized papain were described.The optimum conditions for immobilization were as follows: 20 mg native free enzyme/g of the MCM-41 and 0.75% glutaraldehyde, 2 h at 10-20℃ and pH 7.0. Under these optimum conditions for immobilization, the activity yield [%] of the immobilized enzyme was around 55%. This immobilized papain showed greatly improved thermostability and pH stability compared to its native free enzyme.Ⅲ. Papain immobilization on MCM-48 with the aid of glutaraldehyde was deeply examined on the basis of investigation of papain on MCM-41, and this immobilized papain was well characterized.The optimum conditions for papain immobilization on MCM-4 8 were identical to those for papain immobilization on MCM-41,...
