Ginsenosides has broad physiological activities,the studies of antitumorbiological structure-activity relationships de(?)onstrated the activity of aglycones was stronger than ginsenosides.20(R)-dammarane-3β,12β,20,25-tetrol,a derivative of protopanoxadiol was discovered from the fruits of Panax ginsengand showed stronger anticancer activity than Ginsenoside-Rg3(5-15 times) for the first time.This article applied for the first time the method of Smashing Tissue extraction (STE) to extract the total saponins from stems and leaves of Panaxquin-quefolium L.and optimize the best extraction techniques and acid-degrada-tio-nmethods.The hydrolysate of ginsenosides were isolated and purified by vari-ouschromatographic techniques and 13 compounds were obtained.Among them 9 compounds were identified by spectral and chemical methods as:dammarane-20(22)en-3β,12β,25-panaxatriol(â… ),panaxadiol(â…¡),20(S )-protopanaxadiol(â…¢),20(R)-protopanaxadiol(â…£),20(S)-panaxatriol(â…¤),24(R)-Ocotillol (â…¥),20(R)-protopanaxatriol(â…¦),20(R)-dammarane-3β,12β,20,25-tetrol(â…§), 20(S)-dammarane-3β,6α,12β,20,25-pentol(â…¨).Compoundâ… was discoverd from the total saponins of Panax quinquefolium L.for the first time.The anticancer activities of the compoundsâ…¡,â…£,â…¥,â…§,â…¨were evaluated in seve ral cancer cell lines as human breast cancer cell lines MCF-7,hepatoma carcinoma cell HepG2,human lung cancer cell lines A549,and leukemic cell HL-60.Compoundsâ…§were discovered for the first time from the fruits of Panax ginseng and was identified as the derivate of the aglycone of protopanoxadiol ginsenoside possesing conspicuous anticancer activity.
