Objective:With research method of metabonomics based on NMR, we discuss the characteristics of urine metabonomics of sub-health liver stagnation and spleen deficiency, and discuss macroscopical syndrome of liver stagnation and spleen deficiency from the point of microcosmic metabolic map, which will lay the foundations for objective research on sub-health liver stagnation and spleen deficiency, and discuss application of research method of metabonomics to TCM types of sub-health.Method:choose 12 sub-health patients of liver stagnation and spleen deficiency as experimental group,12 normal persons as normal control group, and 12 sub-health patients of liver stagnation and 12 sub-health patients of spleen deficiency as experimental control group, test the samples of urine by 1H-NMR. After data processing of Pareto scaling, to analyze with PCA method, or adopt PLS method after OSC processing, to study urine metabolites differences between sub-health liver stagnation and spleen deficiency, liver stagnation syndrome, spleen deficiency syndrome and normal person, to look for potential biomarkers of sub-health liver stagnation and spleen deficiency.Result:OSC-PLS (ctr) of four groups of urine'H-NMR spectra revealed different metabolic spectra among sub-health liver stagnation and spleen deficiency dyndrome, liver stagnation syndrome and spleen deficiency syndrome, demonstrated that the metabolic characteristics of the four groups were significantly different. Compared with the urine of normal group,the urine contents of citrate (δ2.54,2.66), TMAO (δ3.26) hippuric acid (δ3.98,7.54,7.58,7.62,7.66,7.82,7.86) were decrease in group of liver stagnation and spleen deficiency, the content and curve relative integral acreage of creatinine (δ3.06,4.06) was increase in group sub-health people; Compared with the liver stagnation dyndrome, the the urine contents of lactic acid (δ1.34,4.14), creatinine (δ3.06,4.06), TMAO (δ3.26), taurine (δ3.26,3.42) increased in group liver stagnation syndrome and spleen deficiency syndrome, the contents and curve relative integral acreage inceased obviously, and the hippuric acid (δ3.98,7.54,7.58,7.62,7.66,7.82,7.86) decreased compared with the normal group; the urine contents of creatinin (5 3.06,4.06) in group liver stagnation syndrome and spleen deficiency dyndrome was more than the group of spleen deficiency, the contents and curve relative integral acreage inceased obviously,but the contents of citrat (δ2.54,2.66), TMAO (δ3.26), hippuric acid (δ3.98,7.54,7.58,7.62,7.66, 7.82,7.86) decreased compared with normal group.Conclusion:Characteristic metabolic products in four groups can be identified by'H-NMR based metabonomics analysis. To analyzed the score chart, loading diagram and original chart could get some potential biomarker of liver stagnation syndrome and spleen deficiency syndrome.The biomaker should be the compound creatinin (δ3.06,4.06) TMAO (δ3.26) hippuric acid (δ3.98,7.54,7.58,7.62,7.66,7.82,7.86) and so on.
