Liver-gistagnation(GY) and-Spleen-qi deficiency(PX) are two relatively independent syndromes. There is a definite relationship in pathogenesis between them. Liver-gi stagnation and Spleen-qi deficiency(GYPX) is a Compound symptom of GY and PX, there is a connection between the three syndromes, But there is little research to discuss. This study base on the " the correlation between formula and syndrome", in order to expound the relevance between GY.PX and GYPX and the interconnectedness between the formula for function of soothing liver and invigorating spleen and the three syndromes. Base on the etiology of Chinese traditional medicine, this study use related pathogeny, the same batch rats to form the three models synchronously. Based on finding of significant changes in the digestive system of former animal model, In the comparative observation of GY.PX and GYPX on the basis of the system changes, then to observe and compare the effects of Chai Shu Si jun Tang on the changes in the digestive system of GY,PX and GYPX in Rat. The dissertation contains both of documental summarization and experimental study. Recent research developments of clinic and experiment about the syndrome of GY.PX and GYPX. The replication and evaluation of three syndromes in rats models and the effective mechanism of the formulae on the changes in the digestive system of three model rats are investigated in the experimental study.Methods:Rats were randomly divided into normal control group, GY group, GY medication group,PX group,PX medication group,GYPX group, GYPX medication group,8-10rats in each group. Adopted by chronic restraint stress, excess fatigue+improper diet and chronic restraint stress+excess fatigue+improper diet to form the model. On the15th day, medicated group rats were given Chai Shu Sijun Tang3.57g·1000g-1·d-1accordingly, model group and normal group were given distilled water for14days. Observe and record all of the emotional behavior in rats, hair color, diet, appearance of urine, and so on. Measured the body weight every week. On the28th day, the10%of chloral hydrate anesthesia animals, abdominal aortic blood, separation test Plasma MTL, GAS, SS, CCK. Detection of the activity of trypsin, lipase and amylopsin in duodenum of each model. Statistical methods:the data were described by (x±SD) and the number of samples compared between groups used one-way ANOVA (one-way ANOVA) test:non-normal data using non-parametric statistical analysis. All data used SPSS17.0statistics software.Experimental results:1. Effects of different Prescriptions on the changes in appearance, weight and the changes in the digestive system.(1) Compared with normal group, the model rats shown the abnormal changes of behavior and the decrease of their weight(p<0.05),PX and GYPX model rats reduced more obviously than GY model rats. Stimulation after two weeks, GY groups starts to irritability, gradually turning indifference and struggle to reduce depression, expression, and companion see diet reduce, defecate is dry. PX groups show that the level of activity reduced, reduced diets and easy to sleep and so on. The GYPX models shown that the level of activity reduced, emotional instability, reduced diet and easy to wake up and so on. After four weeks, the score of activity and emotional in GY rats increased markedly (p<0.05), the score of activity, hair and diet in PX rats increased markedly (p<0.05), the score of activity, emotional, hair and diet in GYPX rats increased markedly (p<0.05); Compared with GY model groups, the score of hair and diet in PX and GYPX rats increased markedly (p<0.05), the score of emotional in: PX model groups reduced markedly (p<0.05). compared with PX models, the score of emotional in GYPX model groups increased markedly (p<0.05).(2) Compared with normal group, the avtivity of the lipase and amylopsin of the GY model groups had improved markedly, D-xylose excretion rate declined markedly (P<0.05); the avtivity of the lipase, amylopsin, pepsin and D-xylose excretion rate declined markedly in PX and GYPX model groups(P<0.05). the three model groups declined markedly on the activity of the trypsin (P<0.01). Compared with GY model groups, the avtivity of the lipase, amylopsin, pepsin declined markedly in PX and GYPX model groups (P<0.01), D-xylose excretion rate have reduce trend.(3) Compared with normal group,the content of motilin, gastrin and cholecystokinin of GY and PX had declined markedly (P<0.05), the content of somatostatin had inproved markedly in PX model groups (P<0.01), the content of motilin, gastrin and somatostatin inproved markedly and cholecystokinin declined markedly in GYPX model groups (P<0.05). Compared with GY model groups, the content of somatostatin inproved markedly and cholecystokinin declined markedly in PX and GYPX models (P<0.05,p<0.01), the content of motilin, gastrin inproved markedly in GYPX models (P<0.01); Compared with PX model groups, the content of motilin, gastrin inproved markedly in GYPX models (P<0.01);2. The effects of Chai Shu Sijun Tang on the changes of three models in Rat.(1) Compared with model groups, the behavior and weight of the three medicated groups had improvement to some degrees, of which GYPX medication group’s weight difference was obvious (P<0.05). the three medicated groups improved markedly on the the level of activity, GY medication groups improved markedly on the emotional state of stability and sleep, PX medication group improved markedly on skin-fur appearance and appetite, GYPX medication group improved markedly on emotional state of stability and sleep,skin-fur appearance and appetite.(2)Compared with each model group, the index of gastrointestinal function of three medicine rats improved to some degrees, the avtivity of the lipase and amylopsin of the GY medication groups had declined markedly (P<0.05), improved markedly on the activity of the trypsin (P<0.01). the avtivity of the trypsin, amylopsin and D-xylose excretion rate of the PX medication groups had improved markedly (P<0.05). the avtivity of the trypsin, lipase,amylopsin, pepsin and D-xylose excretion rate of the GYPX medication groups had improved markedly (P<0.05,p<0.01).(3) Compared with each model group, the content of gastrin of the GY medication groups had inproved markedly (P<0.05);the cholecystokinin of PX medication groups had inproved markedly (P<0.05), declined markedly in somatostatin; the content of cholecystokinin of GYPX medication groups had inproved markedly (P<0.05), declined markedly in motilin, gastrin and somatostatin (P<0.05).The results showed that after two weeks stimulation, the model rats shown the abnormal changes of behavior and the decrease of their weight. The score of activity and emotional in GY rats increased markedly; the score of activity, hair and diet in PX rats increased markedly; the score of activity, emotional, hair and diet in GYPX rats increased markedly; GY rats exist weak intestine absorption and gastric contraction weakened, emptying extended. PX rats shows gastrointestinal digestion, absorption and sports function inhibition in an all-round way. GYPX model rats shows gastrointestinal digestion and absorption of weakened at the same time accompanied by abnormal gastrointestinal movement regulation.The medicated group rats were given Chai Shu Sijun Tang had improvement to some degrees in experiences and weight. Digestive enzymes of GY model rats and part of the gastrointestinal hormones in the blood index has improved, but for some abnormal index has increased. Digestive enzymes of PX model rats and some of the gastrointestinal hormones in the blood index has improved. To change the index almost all of the GYPX rats were significantly improvement.Conclusion:Chronic restraint stress, excess fatigue+improper diet and chronic restraint stress+excess fatigue+improper diet can successfully form the GY, PX and GYPX models. The three model rats represent characteristic to change in appearance, all exist abnormal function of the digestive system, characterized by decreased in different degree of intestinal absorption function. In addition, GY model rats with abnormal secretion.of digestive enzymes and gastrointestinal motor function weakened; PX model rats with digestive enzyme secretion and weakened gastrointestinal motor function; GYPX model rats decreased with gastrointestinal digestive enzyme secretion or dysregulation of gastrointestinal movement. The PX and GYPX model rats showed heavier of gastric bowel function injury. Chai Shu Sijun Tang had better effect on the model of GYPX syndrome.This study provides some objective evidence on the connection between the three syndromes from the changes in the digestive system. Chai Shu Sijun Tang had effect on the function of the digestive system of GY-PX-GYPX models in Rat, but it had better effect on the model of GYPX syndrome indicates that the correlation between prescription and syndrome.
