Shengmai injections derived from the Powder for Restoring Pulse Beat which was recorded in the Medical Origin and Insights, written by Yuansu Zhang. This kind of medicine was first developed by the Medical Science University of West China. This product was made from the sterilized aqueous solution which was obtained via extracting from Radix ginseng Rubra, Ophiopogon japonicus (Thunb.) Ker-Gawl and Schisandra chiensis (Turcz) Ball. Now it is embodied in the fifteenth volume of Ministry of Health Medicine Standard of Prescription Drug Preparations. In 2001, it was ranked in the national protected species of Chinese Medicine. Shengmai injections have been mostly applied to treat different types of cardiopathy, such as myocardial infarction, cardiac shock and infectious shock.In order to study on the therapeutic basis of traditional chinese medicinal preparation shengmai injections, clear pharmacodynamic real Radix ginseng Rubra, Ophiopogon japonicus (Thunb.) Ker-Gawl and Schisandra chiensis (Turcz) Ball influence of shengmai injection, improve the shengmai injection quality standard born, this paper based on the previous studies of the prescription medicine from the shengmai injection which chemical composition and influence on the potency were studied.In the article, fifteen compounds from Ophiopogon japonicus (Thunb.) Ker-Gawl were separated and fourteen chemical structures of which were identified. They are 25(R)-ruscogenin-1-O-α-L-rhamnopyranosyl-(1→2)-[β-D-xylopyranosyl-(1→3)]-β-D-fucopyranoside (MD-2); 25(R)-spirost-5-ene-3β,14α-diol-3-O-α-L-rhamnopyrano syl-(1→2)-[β-D-xyopyranosyl(1→4)]-(3-D-glucopyranoside (MD-3); 25(R)-spirost-5-ene-3β,14α-diol-3-O-α-L-rhamnopyranosyl-(1→2)-β-D-glucopyranoside (MD-4); 25(R)-ophiopogenin-3-O-[α-L-rhamnopyranosyl(1→2)]β-D-xylopyranosyl-(1→4)]-β-D-glucopyranosyl (MD-5); 25(R)-spirost-5-en-3-ol-3-O-[α-L-rhamnopyranosyl (1→2)-β-D-xylopyranosyl-(1→3)]-β-D-glucopyranosyl (MD-6); 25(R)-ophiopoge nin-3-O-a-L-rhamnopyranosyl(1→2)-β-D-glucopyranosyl (MD-8); 25(S)-ruscogenin-1-O-β-D-fucopyranoside3-O-a-L-rhamnopyranoside (MD-9); 25(S)-ruscogenin-l-O-β-D-xylopyranosyl-(1→2)-(3-D-fucopyranoside (MD-11); 25(S)-ruscogenin-1-O-a-L-rhamnopyranosyl(1→2)-β-D-xylopyranoside (MD-12); 25(S)-ruscogenin-1-O-β-D-xylopyranosyl-3-O-a-L-rhamnopyranoside (MD-13); 25(R)-pennogenin-3-O-[a-L-rhamnopyranosyl(1→2)]β-D-xylopyranosyl-(1→4)]-β-D-glucopyranosyl (MD-14); 25(S)-ophiopogenin-3-O-[a-L-rhamnopyranosyl(1→2)]β-D-xylopyranosyl-(1→>3)]-β-D-glucopyranoside (MD-16); 26-O-β-D-glucopyranosyl-(25S)furost-5.20(22)-dien-1β,3β,26-trihydroxy-1-O-β-D-fucopyranosyl-3-O-a-L-rhamnopyranoside (MD-17); 26-O-p-D-glucopyranosyl-(25S)furost-5-ene-1β,3β,22ξ,26-tetrahydroxy-1-O-β-fuc opyranosyl-3-O-a-L-rhamnopyranoside (MD-18); among which MD-11, MD-13, MD-16, MD-17, MD-18 were the new compounds first identified from Ophiopogon japonicus.Six compounds were separated from Schisandra chiensis (Turcz) Ball and five chemical structures among them were identified in this article. They are Schizandrol B (WWZ-1); Lanosterol (WWZ-2); chamigrenal (WWZ-4); Pangeloylgomisin P (WWZ-5); Schizandrol A (WWZ-6).Twelve compounds were separated from Radix ginseng Rubra. They are Ginsenoside Rb1, Ginsenoside Rb2, Ginsenoside Rb3, Ginsenoside Rc, Ginsenoside Rd, Ginsenoside Re, Ginsenoside Rg1, Ginsenoside Rg2, Ginsenoside Rf, Ginsenoside Rh1, Ginsenoside Rh2, Ginsenoside Ro.The determination method of the ginseng saponins from Radix ginseng Rubra, the lignans from Schisandra chiensis (Turcz) Ball and the steroidal saponins from Ophiopogon japonicus (Thunb.) Ker-Gawl in the shengmai injections was established by HPLC for the first time. Literature shows that the ginseng saponins is the effective component of the shengmai injections.Nine kinds of ginseng saponins (including Ginsenoside Rd, Ginsenoside Rb3, Ginsenoside Rb1, Ginsenoside Rb2, Ginsenoside Rc, Ginsenoside Rg2, Ginsenoside Rf, Ginsenoside Re, Ginsenoside Rg1) from Radix ginseng Rubra were detected in the shengmai injections by HPLC.Two kinds of lignans (Schizandrol A and Schizandrol B) from Schisandra chiensis were detected in the shengmai injections by HPLC.The steroidal saponins from Ophiopogon japonicus weren't detected in the shengmai injections by HPLC.The results showed that the contents were quite different between different batches of different manufacturers:Ginsenoside Re:93-120μg.mL-1, Ginsenoside Rg1: 74-100μg.mL-1, Ginsenoside Rf:26-72μg.mL-1, Ginsenoside Rb1:183-340μg.mL-1, Ginsenoside Rg2:11-17μg.mL-1, Ginsenoside Rc:120-178μg.mL-1, Ginsenoside Rb2: 86-195μg.mL-1, Ginsenoside Rb2:11-17μg.mL-1, Ginsenoside Rd:37-59μg.mL-1, Schizandrol A:4-15μg.mL-1, Schizandrol B:0.4-1.6μg.mL-1.According to the results of the study prepared by the aforementioned respectively, three kinds of injections (the injections from nine ginseng saponins and two lignans mixed, the single injections from nine ginseng saponins mixed and the single injections from two lignans mixed) and control shengmai injection scenedesmus born for a comparative study on after myocardial infarction rats to heart failure of protection. The results show that the obviously protection effect of the injection from nine kinds of ginseng saponins and two kinds of lignans mixed, the single injection from nine kinds of ginseng saponins mixed and the shengmai injections scenedesmus is fairly. Only the ginseng saponins may be the main effective component of the shengmai injections.This thesis research suggests the ginseng saponins from Radix ginseng Rubra of the shengmai injections has the positive effect of protecting cardiac muscle. This thesis provides scientific basis on the therapeutic basis and the second development of the shengmai injections.
