Background: Renal blood perfused organs are sensitive to ischemia and ischemia-reperfusion. Renal ischemia-reperfusion injury can lead to acute tubular necrosis oracute renal insufficiency. At present, studies suggest that the pathophysiology of renal (I/R), is very complex, involving inflammation, apoptosis and oxidative stress injury.Tanshinone is an effective active ingredient of Salvia miltiorrhiza, has anti-ischemichypoxia, improve microcirculation, inhibition of platelet aggregation and otherfunctions and antithrombotic effects, which tanshinone â…¡ A (Tanshinone â…¡ A Tanâ…¡ A) Tanshinone in the highest concentrations of active ingredients. Currently noreports of Tanshinone â…¡ A treatment of kidney (I/R).Objective: This study Tanshinone â…¡ A study to evaluate its protective effect onrenal ischemia-reperfusion injury, and Tanshinone â…¡ A mechanism through whichrenal ischemia-reperfusion injury from a protective effect.Methods: Male SD rats were40, weight200grams and250grams, were randomlydivided into sham operation group (C), renal ischemia-reperfusion group (IR),tanshinone â…¡ A administration group (T1), tanshinone â…¡ A administrationgroup2(T2), groups of10rats.Part I: observation groups ultrastructure, morphology and renal function thechange. Group of electron microscope, HE staining, detection of Cr.Part II: observation of the impact of oxidative stress pathway of tanshinone II A(Tanshinone II A Tan II A) on renal ischemia reperfusion injury. Were detected by MDA content, SOD.GSH-Px and CAT activity.Part III: observe the apoptosis pathway of tanshinone II A (Tanshinone II A Tan II A)on renal ischemia reperfusion injury. TUNEL method to detect apoptosis of cells in eachgroup, to detect groups of bcl-2and bax expression level changes.Results:1A subject of electron microscopy results show that: T1and T2group comparedwith IR group, The ultrastructure significant improvement of Tan II A can reduce therenal ultrastructural damage.2The two topics under the light microscope was observed: T1and T2groupcompared with IR group, significant improvement of tissue morphology, Paller's scoredecreased significantly (P <0.01) to prove that Tan II A can alleviate the injury of renaltissue morphology.3Topics of Cr test results: T1and T2Cr levels were significantly lower than the IRgroup (P <0.01), proof of Tan II A can reduce the renal dysfunction.4Compared with IR group, T1and T2group of SOD, CAT and GSH-Px activitywas significantly increased MDA levels were significantly reduced (Table2.1-2.4),indicating that Tan II A, due to increased oxygen free radical scavenging enzymes ofSOD, CAT, and GSH-Px activity, which cleared the number of oxygen free radicalswhen the IRI, so that the lower level of oxygen free radicals mediated MDA levelsdecreased.5Compared with IR group, T1and T2apoptotic index was significantly lower (P<0.01), confirmed the anti-apoptotic effects of Tan II A, indicating that Tan II A canreduce apoptosis in renal ischemia-reperfusion.6Compared with IR group, T1and T2group of Bcl-2mRNA and proteinexpression increased (P <0.01, P <0.01), Bax mRNA and protein expression decreased(P <0.01, P <0.01), T1and T2group Bax/Bcl-2protein ratio (Figure3.11) reduction,reduced T1and T2apoptosis, in line with with the TUNEL assay (Fig.3.2-3.4).Conclusion:1Tan II A can reduce the injury of the kidney ultrastructure, morphology and function, has a protective effect on renal ischemia reperfusion injury.2Tan â…¡ A num berofclearIR Ithrough increased SO D,C A T and G SH-Px activity,oxygen free radicals, oxygen free radical levels decreased, in order to play itsantioxidant effects.3Tan â…¡ A protective effecton ratkidney IR Iand its antioxidanteffectis related to.4Tan II A by increasing the Bcl-2mRNA and protein expression, reducing BaxmRNA and protein expression, thereby reducing the Bax/Bcl-2protein ratio, andinhibition of apoptosis.5Tan â…¡ A through inhibition of apoptosis,play a protective effect on rat kidneyIRI.
